I believe GameStop regional or district managers can go, too.

They are making it harder and harder for that to happen.

Question: Other than the Big 3, do we know who is giving conferences?

Just about every big company - Capcom, EA, Ubisoft, Square Enix, etc have conferences.

So, I wonder if Disney and Square being there might mean something

Both Disney and Square are there EVERY year. Doesn't mean much.

IGN and GameSpot will livestream the conferences, as always. So will G4TV.
And GameTrailers.

So for that contest...they want us to just film the opening question...like "Dear Playstation, blah blah blah blah blah blah blah" ? That's weird.

And yet so simple. I guess if you win not only do you get to go to E3 but they might use your idea for an actual Kevin Butler PS3 commercial. I don't know. But it's an easy shot at getting in E3.
